This^^^ If this was after the whole jada cheating/him crying thing then yes. He is not alpha. Especially since he was laughing at the joke at first, until he sees jada is not happy with it.

He did the honorable thing in defending his wife, just went about it wrong. Should've "confronted" him at a later time that night when the camera's weren't rolling. In the eyes of many he just came across as a fool.

The key was he went up there only after seeing his "wife" upset at the joke. He looked about his whipped as one possibly could. Not to mention he did this on live TV knowing that Rock wasn't going to retaliate in any serious fashion OR if he did there would have been 50 people rushing both of them to put a stop to it.

As much as I like Will Smith as an actor, and seems like a cool guy to hang out with I lost a bit of respect for the whole Jada thing a while back. He is what I would have considered a high-value man, no reason why he needs to conform to Jada and her antics.
As for the altercation, slapping another man is not necessarily alpha. Especially because he is protecting someone who completely disrespect him and emasculated him in public.
