That is an insult. Not a good way to pick up a girl. Neg hits are not meant to offend the other person. The neg hit is used to break her B1tch shield. that transexual joke offended her and probably turned her off. A neg hit is more like "You have nice eyes, but you wear way too much eye liner" or "That's a nice outfit, but red is not your color."

That being said, 19 out of 20 times, what you just said will be an insult. And if you have to ask, it's an insult.
