Okay, so i'm a sophomore, and my sister is a freshman. At lunch there is this one girl that I like to mess with, with my friends. She's cute, and there's no harm in it, but this girl goes off to talk to my sister about me.
She pretty much asked my sister why I don't like her or something like that.
I really don't know how to interpret this.
Does this mean that she's inerested in me or what?

It doesn't mean she's necessarily interested in you. Did you hear her ask it? The tone of her voice? She may have been just having fun like, "Oh my God, why doesn't he like me? Lol" or upset, like, "I don't understand why he doesn't like me...." In any case don't take this as a sign that she's interested, even though she might be. She could just view you as a friend.

I don't know how to interpret this really. I mean, should i call her? I just don't want this to be a joke or something.
